Generally, emulsion can be prepared using a two-syringe method. Two syringes are connected with a connector. Each syringe contains equal volume of antigen solution and adjuvant (generally Freund complete adjuvant : CFA). With two 1 ml syringes, 0.3- 0.5ml volume each can be used. Antigen solution can be 2 mg/ml and CFA can be 1 or 2 mg/ml M tuberculosis in mineral oil. They are mixed 100 strokes each, then repeat 3 times. The emulsion quality is very important to induce immune response against the antigen. The antigen with M.tuberculosis must be released gradually in 2-3 weeks. The quality can be checked easily. Remove the connector and attach a needle. Then make a drop of emulsion on water in a beaker. The drop must keep its shape without dispersing. This promise that the emulsion was formed as a water in oil structure (1).
